興趣:

磷酸水解酶PP2A在近來已被認為扮演抑制腫瘤形成的角色。PP2A完全酶是由三種次單元所組成,包括了催化次單元C、結構次單元A、及多變的調節次單元B。目前至少有四種B調節次單元家族被發現,而PP2A的多元組合及功能是由於多變的B次單元所造成。我們探討PP2A在抑制腫瘤形成的分子機制,也致力於探討B次單元所扮演的細胞角色。近年來我們也發現PP2A可能扮演促進癌症轉移的証據,目前致力探討其可能的機制及在動物實驗上驗證。我們建立了合併雙分子螢光互補作用BiFC及螢光共振能量轉移FRET的光學影像實驗方法來探討在活細胞中直接觀察PP2A由ABC次單元組合的三聚體及調節機制。

我的實驗室的研究方向主要有:

  1. 探討磷酸酶PP2A在細胞癌化的角色。目前著重在探討帶有B56 gamma3 調節次單元的PP2A在大腸直腸癌中腫瘤轉移的角色及調節細胞癌化的機制。
  2. 以應用合併雙分子螢光互補作用(BiFC)及螢光共振能量轉移(FRET)方法探究在細胞中PP2A複合體的變化,並藉由此方法來篩選新穎的PP2A複合體的調節分子。
  3. 針對探討PP2A與p70S6K之間交互作用的機制來研發癌症新藥。
  4. 探討PP2A-B55δ在大腸直腸癌中的角色及在腫瘤免疫調節的角色。
