HBV and HCV infections and their severe complications with cirrhosis and hepatocellular carcinoma are ones of major
issues in medicine. Hosts and viruses play distinctly important roles on mechanism of viral invasion, disease progression, and treatment response. Our laboratory focuses on the interaction of immunologic and genetic characteristics between host and viral genomes, via integrating clinical and basic researches.


1. Hepatocellular carcinoma-associated single-nucleotide variants and deletions identified by the use of genome-wide high-throughput analysis of hepatitis B virus: We initiate NGS researches in hepatobiliary diseases. Based on our experiences on studying HBV and hepatocellular carcinoma, we are enthusiastic and confident to accomplish this research project which is of great significance from either academic or medical angle.

2. Clinical significance of serum hepatitis B virus RNA in chronic hepatitis B patients receiving long-term nucleos(t)ide analogue therapy, with further investigation of hepatitis B virus covalently closed circular DNA biology: Current anti-hepatitis B virus (HBV) therapies rarely achieve a cure of chronic HBV infection due to the presence of covalently closed circular DNA (cccDNA). Serum hepatitis B surface antigen (HBsAg) and hepatitis B core-related antigen (HBcrAg) reflect transcriptional activity of intrahepatic cccDNA and are useful biomarkers during and after cessation of antiviral therapy. Recent studies showed that serum HBV RNA is associated with treatment responses and viral rebound after withdrawal of treatment. However, the kinetics of HBsAg, HBcrAg, and HBV RNA during and after cessation of antiviral therapy remained to be determined. This study will help us to understand the impact of nucleos(t)ide analogues treatment on HBV life cycle and cccDNA biology.

3.Identification of the diversity of IgG glycosylation and its immunomodulatory effects in HBV-related liver diseases:
In this project, we will study the correlation between serum IgG glycosyl patterns and the development of HBV-related liver diseases. In addition, we will investigate the possibility of IgG glycosyl restoration during the remission of liver diseases and the reversal of cirrhosis, thus to identify the specificity of IgG glycoforms and progression of hepatic diseases. Finally, immunomodulatory effects of IgG glycovariants on macrophages phagocytic activities will be demonstrated. We expect to employ IgG glycomes on clinical diagnosis and be a prognostic marker of HBV-related liver diseases.
