  One of the research interests in my laboratory is tumor metastasis. Metastasis is the spread of cells from the primary tumor site to distal organs, and is the major cause of death in cancer patients. We focus on neuroblastoma, which is a common childhood solid tumor arising from the neural crest that develops into the sympathetic nervous system. Because of the embryonic nature, the genetics and biology of neuroblastoma likely differ from adult tumors. In fact, we have demonstrated that nucleoside diphosphate kinase A (NDPK-A) acts as a metastasis promoter in neuroblastoma, which contrasts with the metastasis-suppressing role of NDPK-A in adult tumors such as breast carcinoma. Furthermore, metastasis-associated amplification and mutation of the nm23-H1 gene that encodes NDPK-A alters neuronal differentiation, cell survival, adhesion and migration. Based on our results from yeast two-hybrid and DNA microarray experiments, we are investigating candidate proteins and genes in the NDPK-A pathway that participate in neuronal differentiation, cell adhesion/migration, and tumor metastasis.

   Another research interest of my laboratory is cancer prevention, which targets the DNA mismatch repair system (MMR). Cancer is a genetic disease resulting from not only chromosomal instability but also microsatellite instability. The MMR system maintains genome integrity by eliminating replicative errors that frequently occur in microsatellites. Mutational inactivation of the MMR function leads to microsatellite instability (MSI). The MSI phenotype has been detected in human cancers having MMR gene mutations and in inflammatory diseases caused by infections and other factors. We are developing cell and mouse models to study microsatellite instability and to screen cancer-preventing compounds.
